肝素层层自组装修饰猪肝去细胞化支架材料的抗凝血性能研究

摘要:目的:通过对猪肝去细胞化材料进行肝素层层自组装修饰改善其作为肝脏组织工程支架材料的抗凝血性能。方法:对获取的猪肝去细胞化材料进行进行层层自组装(layer-by-layer elf-assembly technique, LBL)肝素化修饰,检测其肝素化效果及抗凝血性能。结果:猪肝去细胞化材料经肝素化修饰后材料内表面有肝素结合,并能缓慢释放。肝素修饰后的材料各项抗凝血性能明显改善;同时,溶血试验表明修饰材料符合生物材料的溶血性要求。结论:LBL 肝素化修饰能显著改善猪肝去细胞化材料体外抗凝血性能。

Anticoagulant ability of layer-by-layer self-assembly technique heparinized decellularized porcine liver scaffolds

Abstract: Objective:Enhance the anticoagulant properties of the decellularized porcine liver
biological materials as scaffold for tissue engineering research via heparinized process. Methods: he decellularized porcine liver scaffolds were immobilized with heparin through layer-by-layer self-assembly technique (LBL). The effect of heparinization and anticoagulant ability were tested.Results:The results show that the scaffolds had been successfully immobilized with heparin. The Heparinized scaffolds gained better performance of the anticoagulant than control scaffold.
Hemolysis test revealed that the heparinized material did not lead to hemolysis. Conclution:The decellularized porcine liver for tissue engineering acquired a good effect of anticoagulant ability in vitro via LBL heparinized technique.
