Gli相关基因调控网络及其在癌症发生发展中的作用及机制

摘要:Gli(Glioma-associated oncogene homolog)是重要的核转录因子之一,研究表明,其不仅接受经典Hedgehog信号通路的调节,同时受多种其他非经典信号通路如TGFβ10 /SMAD,EGFR等信号通路的调控。Gli作为信号通路的终端效应因子,可以直接调控多种目的靶基因的转录表达。Gli及其靶基因网络的异常可直接影响细胞,进而导致组织及个体的生物学性状的改变。近年来,随着组学技术的发展,通过ChIP-chip、基因芯片DNA测序分析等技术,为发现转录因子Gli的直接靶基因或效应靶基因,深入了解其相应调控网络提供了更多的机会。本文就Gli调控的基因网络及其在癌症发生发展中的作用及机制,并可能15 带来的肿瘤治疗、诊断等前景作一阐述。

Abstract: Gli(Glioma-associated oncogene homolog)is one of the important nuclear factors. Not only the Hedgehog/Gli signaling pathway, but also the non-canonical signaling pathway (like TGFβ/SMAD,EGFR signaling pathway) can regulate Gli. As the end of effector, Gli can directly 30 regulate the transcription of many target genes, further influence the biological changes of cell,tissue and individual. Recently,as the development of Omics technology. This review illustrates the control gene web of Gli, and the function and mechanism related to tumorigenesis and development, the future tumor therapy solution.
