 大鼠脑基底动脉最适初始张力的确定

摘要:目的:确定SD大鼠脑基底动脉收缩的最适初始张力。方法:将大鼠脑基底动脉制成1~2 mm长的环段,用两根钢丝悬挂于Myograph 恒温浴槽中。将IC1/IC100值设置为0.6~1.0,记录其相对应的初始张力。在不同的初始张力下,记录60 mM KCl诱导的收缩力,研究诱导的最大收缩力与初始张力的关系。结果:随着IC1/IC100值从0.6至1.0,脑基底动脉初始张力逐渐从0.47±0.01 mN/mm 增加到2.68±0.03 mN/mm,对IC1/IC100和初始张力进行相10 关性分析,相关系数为0.95,表明二者正相关。KCl诱导的收缩力先随IC1/IC100值升高而升高,当IC1/IC100值为0.9、初始张力为1.63 mN/mm时,KCl诱导的收缩力达到最大,其值为6.51±0.05 mN/mm,明显高于IC1/IC100在0.6和0.7时的收缩力(P<0.05)。此后,随着IC1/IC100值的增加,KCl诱导的收缩力不再增加,反而呈下降趋势。结论:大鼠脑基底动脉环的最适初始张力为1.63±0.01 mN/mm。

An optimal initial tension of rat basilar artery in wire myography

Abstract: Objective: The present study was to determine the optimal initial tension in rat basilar artery. Methods: Rat basilar arteries were cut into approximately 1~2 mm, threaded on two thin wires (40 μm in diameter), and mounted in myograph baths. Normalization procedure was carried 25 out. IC1/IC100 was set from 0.60 to 1.00, and the initial tensions were recorded. KCl 60 mM was added into the baths to induce a contractile tension. Results: The initial tension of basilar artery increased from 0.47±0.01 mN/mm to 2.68±0.03 mN/mm as the value of IC1/IC100 increased from 0.6 to 1.0. The correlation coefficient between IC1/IC100 and initial tension was 0.95. At first, the contractile tension was elevated with the value of IC1/IC100 and initial tension increasing. When 30 the IC1/IC100 and initial tension reached to 0.9 and 1.63 mN/mm, the contractile tension reached its maximum value, 6.51±0.05 mN/mm. Then the contractile tension started to decrease as IC1/IC100 or initial tension increased. The contractile tensions in IC1/IC100 value of 0.9 was significantly higher than in the IC1/IC100 value of 0.6 and 0.7 (P<0.05). Conclusion:.The most appropriate initial tension is 1.63±0.01 mN/mm.
