摘要:目的 分析PD-1 在慢性HCV 感染者CD4+T 细胞亚群的表达情况,以及PD-1 表达水平与HCV 血浆病毒载量和酶学指标的相关性。 方法 采用流式细胞术对19 例慢性HCV 感染者和17 例健康对照者进行了外周血CD4+T 细胞亚群和表型分析,及各细胞亚群PD-1 表达水平的测定。同时将细胞亚群的比例,PD-1 表达水平与HCV 病毒学指标及酶学指标进行了关联性分析。 结果 慢性HCV 感染者Naïve T 细胞明显低于健康对照者,而TEM 细胞和终末效应T 细胞明显高于健康对照者。PD-1 在慢性HCV 感染者中各CD4+T 细胞亚群和总CD4+T 细胞中的表达水平升高。慢性HCV 感染者PD-1 表达水平升高与HCV 病毒学指标及酶学指标之间无明显相关性。 结论 本研究系统分析了HCV 长期慢性感染者体内外周血CD4+T 细胞亚群分布特征。结果显示分析患者外周血CD4+T 细胞表面成熟和活化抑制标志对评价宿主细胞免疫水平提供了一个很好的参考指标。

The study of PD-1 expression on different CD4+T cell subsets in chronic HCV infected patients

Abstract: Objective To evaluate the PD-1 expression on different CD4+T cell subsets in chronic HCV infected patients, and investigate the correlation of PD-1 abnormal activating expression on 25 CD4+T cell with HCV virological and enzymological indicators. Methods A study of the percentage of CD4+T cell subsets and MFI of PD-1 expression on total CD+T cells and its subsets were performed by multi-parameter flow cytometry in 19 chronic HCV infected individuals and 17 healthy control volunteers. Additionally, we studied the correlations between PD-1 expression and HCV Virological and enzymological indicators. Results The percentage of Naïve T cell subsets was lower but TEM and Effector T cell subsets were higher in 19 chronic HCV infections compared to healthy control. The level of PD-1 in total CD4+T cells and four CD4+T cell subsets
increased dramatically in chronic HCV infections. There was no significant difference between the upregulation of PD-1 and the HCV virological and enzymological indicators. Conclusion The distributional pattern of peripheral CD4+T cell subsets and the characteristics of PD1 expression on CD4+ T cells in chronic HCV infected patients were investigated. The finding indicated that it was meaningful to evaluate the status of host cellular immune response through monitoring maturation (CD45RA and CD27) and inactivation marker PD-1 on peripheral CD4+T cells and its subsets in chronic HCV infected patients.
