摘要: 目的:观察RNAi 抑制前列腺癌PC-3 细胞中的PHB 表达对细胞增殖、凋亡影响,探讨PHB 对人前列腺癌的可能作用机制。方法:SiRNA 基因沉默PC-3 细胞中PHB 基因, Annexin V/PI 双染和流式细胞计数观察NM 和CDM 培养下及分别给予血小板源性生长因子PDGF 或EGF10 刺激的前列腺癌凋亡细胞数目差别 。结果:流式细胞仪(FCM)检测发现,在siRNA 抑制PC-3细胞中的PHB 后,CDM 实验组细胞凋亡率显著增加,同NM 对照组比较有显著性差异(p<0.05 );在CDM 组给予PDGF 或EGF 后,细胞凋亡率升高,与NM 对照组相比较有显著性差异(p <0.05 )。结论:低胆固醇培养条件下前列腺癌细胞生长抑制,给予生长刺激因子促进细胞凋亡,低胆固醇抑制前列腺癌PC-3 细胞体外增殖活性可能同PHB 表达升高有关。

25 Abstract: Objective:To observe the effects of RNAi inhibit PHB on cell growth,and apoptosis in Pc-3 cells.Explore the possible molecular mechanism of PHB on human prostate cancer.Methods:SiRNA knockdown PHB in PC-3 cells and treated cells with NM or CDM added/or not PDGF /EGF.Apoptosis was determined by Annexin V/PI double staining technique and flow cytometry.Results:Annexin V/PI double staining and flow cytometry discovered that after using siRNA knockdowned 30 PHB in PC-3 cells,PC-3 cells apoptosis rate of cultured in CDM was significantly higher than cells cultured in NM(p<0.05).Addred PDGF or EGF into CDM can upon apoptosis rate(p<0.05 ).Conclusions:Cholesterol depleted conditions can induce apoptosis in PC-3 cells.Cholesterol depleted conditions add growth factors can promote apoptosis in PC-3 cells.
