摘要:目的 探讨组织蛋白酶C(cathepsin C)的表达与炎性因子刺激之间的关系,明确在炎症条件下cathepsin C 表达增强的诱导因素。方法 利用LPS 和炎性因子IL-1β,IL-6 刺激原代培养的小胶质细胞和小胶质细胞株BV2,利用比色法检测cathepsin C 酶活性的改变,ELISA 方法检测小胶质细胞cathepsin C 的表达和释放。结果 在炎性刺激后,小胶质细胞中cathepsin C 的活性上调,cathepsin C 蛋白的表达及释放增强,并与刺激强度之间有显著的剂量依赖关系。结论 LPS 和炎性因子IL-1β,IL-6 诱导cathepsin C 的表达并增强其活性。

Inflammatory factors increase microglial cathepsin C expression and activity

Abstract: In this paper, the relationships between cathepsin C and inflammatory cytokines as well as factors inducing cathepsin C expression in inflammation was investigated. Primary microglia and cell line BV2 were treated with LPS and pro-inflammatory cytokines IL-1β, IL-6. Colorimetry as used to examine enzymatic activity of cathepsin C;ELISA was used to determine the expression and secretion of cathepsin C. Results showed that in response to inflammatory
stimulation, cathepsin C expression, secretion and activity in microglia was increased in dose-dependent manner. These data indicate that LPS and pro-inflammatory cytokines IL-1β, IL-6 induce the expression of cathepsin C and up-regulate its activity in microglia.
